ABSTRACT

Abstract Urban agriculture is very common in the cities, as Rio de Janeiro and adjacencies, being important as economic activity. However, this practice can create appropriate conditions for establishment and maintenance of life cycle of many parasites. Lymneid snails can act as intermediate hosts of many trematodes species, including those that infect humans and wild and domestic. In the present study, the snail Lymnaea columella was collected from an urban watercress plantations and the morphology of cercariae released by them was analyzed by light and scanning electron microscopy. The specimens were identified as xiphidiocercariae, belonging to the Cercariae Ornatae group, of the order Plagiorchiiformes. Histological examination of the digestive gland of the infected snails revealed the presence of developing sporocysts, with hemocitary infiltration and metaplasia in the epithelial tissue.


Resumo Agricultura urbana é muito comum nas cidades, como Rio de Janeiro e adjacências, sendo uma importante atividade econômica de subsistência. No entanto, esta prática pode criar condições adequadas para o estabelecimento e manutenção do ciclo de vida de muitos parasitos. Os moluscos limneídeos podem atuar como hospedeiros intermédios de muitas espécies de trematódeos, incluindo aqueles que infectam humanos e animais selvagens e domésticos. No presente estudo, o molusco Lymnaea columella foi coletado de plantações urbanas de agrião e a morfologia das cercárias liberadas por eles foi analisada por microscopia eletrônica de varredura leve e de varredura. As larvas foram identificadas como xifidiocercárias, pertencentes ao grupo das Cercariae Ornatae, Ordem Plagiorchiiformes. Análises histológicas da glândula digestiva dos moluscos infectados evidenciaram esporocistos, com formação de granuloma e metaplasia do tecido epitelial.

ABSTRACT

Background: Epigenetic modifications are key factors modulating the expression of genes involved in the synthesis of phytochemicals. The knowledge of plant epigenetic and genetic variations can contribute to enhance the production of bioactive compounds. These issues have been little explored thus far in Rorippa nasturtium var. aquaticum L. (watercress), an edible and medicinal plant. The aim of the current study was to determine and compare the phenolic composition and epigenetic and genetic variations between wild and cultivated watercress. Results: Significant differences were found in the quantitative phenolic composition between wild and cultivated watercress. The eight primer combinations used in the methylation-sensitive amplification polymorphism (MSAP) method revealed different epigenetic status for each watercress type, the cultivated one being the most epigenetically variable. The genetic variability revealed by the EcoRI/MspI amplification profile and also by eight inter-simple sequence repeat (ISSR) primers was different between the two types of watercress. The results of the Mantel test showed that the correlation between genetic and epigenetic variations has diminished in the cultivated type. Cluster analyses showed that the epigenetic and genetic characterizations clearly discriminated between wild and cultivated watercress. Conclusions: Relevant chemical, epigenetic, and genetic differences have emerged between wild and cultivated watercress. These differences can contribute to fingerprint and develop quality control tools for the integral and safety use and the commercialization of watercress. The richness of epialleles could support the development of tools to manipulate the watercress epigenome to develop high bioproduct­producing cultivars

Introduction: the Eclipta alba is kwoned as ``agrião do brejo´´ in Brazil. Etnomedical studies consider it as antinflamatory and analgesic plant. Objective: To determine the excitation`s threshold of nociceptive system in mice trated whith diferents doses of hydroalcoholic stratum, 30% of Eclipta alba dead roots. Methodology: were used 42 Swiss mice bewteen 8-10 weeks old, constant ambiental temperature of 25ºC and water / food "ad libitum". The analgesic activitiy was determined by the ``hot plate´´ test and the analgesic indicator was the time`s reaction of each animal. Result and Discussion: analgesic action of hydroalcoholic stratum, 30% of Eclipta alba dead root, was confirmed at 60, 90 and 120 minutes. This activity was similar as paracetamol analgesic action (12.5 mg.kg-1). The analgesic efect of paracetamol was increasing for 150 minutes. The prostaglandins inhibition synthesis may be a cause of the analgesic action. Antioxidant effect of phenol and polyphenols compounds presents in the hydroalcoholic stratum, 30% of Eclipta alba dead root and the inhibition of phospholipase A2 by flavonois compounds presents in the stratum too, may be explain the results of this paper, througt the low level of prostaglandins. Conclusion: the hydroalcoholic stratum, 30% of Eclipta alba dead root (500 and 250 mg kg-1) and paracetamol (12.5 mg.kg-1) showed a similar analgesic activity bewteen 60 and 120 minutes


Introdução: a Eclipta alba, conhecida como "agrião do brejo" é uma planta nativa do Brasil, China e Austrália, os estudos etnomédicos reportam entre outras ações a antinflamatória e analgésica, e é esta última, a atividade abordada neste trabalho. Objetivo: verificar a ação analgésica do extrato fluído hidroalcoólico de raíz de Eclipta alba a 30% em camundongos Swiss. Metodologia: utilizou-se 42 camundongos Swiss, machos, entre 8 e 10 semanas de vida, submetidos a temperatura constante e água e comida ad libitum. A analgesia foi determinada através do modelo do "prato quente" e o indicador da variável foi o tempo de reação ao calor. Resultados e Discussão: foi comprovado que aos 60, 90 e 120 minutos de prova, a atividade analgésica do extrato fluído foi similar à atividade analgésica do paracetamol, dose de 12.5 mg.kg-1 de massa corporal, o efeito deste último foi crescente ao longo dos 150 minutos de experimento, evidenciando sua potencia analgésica. Tais efeitos são provavelmente decorrentes da inibição da síntese de prostaglandinas (principalmente da serie "E") causando, portanto, aumento do limiar de excitação das terminações nervosas livres e consequentemente, analgesia. Provavelmente, componentes do extrato hidroalcoólico de raíz de Eclipta alba a 30% causaram inibição das ações da fosfolipase A2 refletindo em menor liberação e transformação do ácido araquidónico em prostaglandinas. Conclusão: doses de 250 e 500 mg.kg-1de massa corporal de extrato fluído, mostraram efeito analgésico comparável ao paracetamol na dose de 12.5 mg.kg-1 de massa corporal, tendo em conta o modelo animal e tempos de provas utilizados neste trabalho

Watercress (Nasturtium officinale, Cruciferae; W. Aiton) is a vegetable widely consumed in our country, with nutritional and potentially chemopreventive properties. Previous reports from our laboratory demonstrated the protective effect of watercress juice against DNA damage induced by cyclophosphamide in vivo. In this study, we evaluated the in vivo effect of cress plant on the oxidative stress in mice. Animals were treated by gavage with different doses of watercress juice (0.5 and 1g/kg body weight) for 15 consecutive days before intraperitoneal injection of cyclophosphamide (100 mg/kg body weight). After 24 h, mice were killed by cervical dislocation. The effect of watercress was investigated by assessing the following oxidative stress biomarkers: catalase activity, superoxide dismutase activity, lipid peroxidation, and glutathione balance. Intake of watercress prior to cyclophosphamide administration enhanced superoxide dismutase activity in erythrocytes with no effect on catalase activity. In bone marrow and liver tissues, watercress juice counteracted the effect of cyclophosphamide. Glutathione balance rose by watercress supplementation and lipid oxidation diminished in all matrixes when compared to the respective control groups. Our results support the role of watercress as a diet component with promising properties to be used as health promoter or protective agent against oxidative damage.


El berro (Nasturtium officinale, crucíferas; W. Aiton) es una hortaliza ampliamente consumida en nuestro país, con valor nutricional y propiedades potencialmente quimiopreventivas. En trabajos previos demostramos que el jugo de berro tiene efecto protector in vivo contra el daño del ADN inducido por ciclofosfamida en tejidos del ratón. En el presente trabajo evaluamos, también in vivo, los efectos del jugo sobre el estrés oxidativo en diferentes tejidos del ratón. Los siguientes biomarcadores fueron investigados: actividad de superóxido dismutasa, actividad de catalasa, peroxidación lipídica y balance de glutatión. Los animales fueron tratados con diferentes dosis de jugo (0.5 y 1 g/kg de peso corporal) por alimentación forzada durante 15 días consecutivos antes de la inyección intraperitoneal con ciclofosfamida (100 mg/kg). La ingesta de berro antes de la administración de ciclofosfamida mejoró la actividad de superóxido dismutasa en los eritrocitos sin efecto sobre la actividad de la catalasa. En médula ósea e hígado, el jugo de berro contrarrestó el efecto deletéreo de la ciclofosfamida. En todas las matrices, el balance de glutatión fue mayor y la oxidación de lípidos menor que los valores encontrados en los grupos control. Nuestros resultados demuestran que el berro es un componente de la dieta con propiedades prometedoras como promotor de la salud o como agente protector contra el daño oxidativo.

ABSTRACT

O cultivo hidropônico de hortaliças folhosas vem crescendo acentuadamente nos últimos anos, no entanto, pouco se sabe sobre sua composição química, o que é de extrema importância, tendo em vista a mudança de hábito alimentar de uma fração considerável da população brasileira que busca alimentos de qualidade. Por essa razão, as culturas de agrião, chicória, rúcula e alfaces americana (cv. Lucy Brown) e lisa (cv. Regina) foram produzidas em hidroponia, sistema NFT, buscando-se avaliar o rendimento, teor de nitrato e composição centesimal. O experimento foi conduzido em casa de vegetação, na Universidade Federal de Santa Catarina, Florianópolis (SC), no período de maio a julho de 2004, sob delineamento inteiramente casualizado com 4 repetições. O espaçamento utilizado foi de 25 cm x 25 cm para as culturas do agrião, chicória e alface (cv. Lucy Brown e cv. Regina) e de 5 cm x 5 cm para a rúcula. A alface cv. Regina e a chicória apresentaram maior número de folhas por planta. O agrião apresentou menor teor de água e maior fitomassa seca da parte aérea. Maior incremento de fitomassa fresca foi obtido na chicória, alface cv. Lucy Brown" e cv. Regina. Maiores teores de lipídios, proteína, resíduo mineral, carboidratos, valor calórico, nitrato e de fibra foram obtidos no agrião. A rúcula apresentou menores valores para as variáveis fitomassa fresca e seca da planta inteira, da parte aérea e da raiz, e número de folhas por planta. Todas as culturas apresentaram boa aparência visual, baixo valor calórico e teores de nitrato adequados ao consumo humano.


The hydroponic cultivation of vegetables has increased markedly in recent years, however, little is known about its chemical composition, which is of extreme importance in view of changing food habits of a considerable fraction of the population seeking food quality. For this reason, cultivation of watercress, chicory, rocket and lettuce American (Lucy Brown) and smooth (cv. Regina) were grown in hydroponics, NFT system, aiming to evaluate the yield, nitrate content and chemical composition. The experiment was conducted in a greenhouse at the Universidade Federal de Santa Catarina, Florianopolis city in the Santa Catarina State (Brazil), from may to july 2004 under a completely randomized design with four replications. The spacing used was 25 x 25 cm for the crops of watercress, chicory and lettuce (cv. Lucy Brown and cv. Regina) and 5 x 5 cm to the rocket. The traits were: number of leaves, nitrate content and chemical composition of vegetables. The lettuce cv. Regina and chicory had higher number of leaves per plant. The watercress had lower water content and higher dry mass of shoots. The largest increase in fresh weight was obtained in chicory, lettuce, cv. Regina. Higher levels of lipids, protein, ash, carbohydrates, calories, fiber and nitrate were obtained from the watercress. The rocket had lower values for the variables fresh and dries the whole plant, shoot and root and leaf number per plant. All cultures showed good visual appearance, low calorie and nitrate levels suitable for human consumption.

ABSTRACT

A utilização de extratos vegetais vem se tornando uma alternativa importante para a prevenção de doenças periodontais. Este trabalho objetivou desenvolver uma formulação de enxagüatório bucal, contendo, em associação, extratos hidroalcoólicos de Rosmarinus officinalis, Plantago major, Tabebuia impetiginosa, Achillea millefollium e Nasturtium officinale; avaliar sua composição farmacognóstica e sua atividade antibacteriana, como também da fórmula proposta. Foram realizados estudos de pré-formulação e análises farmacognósticas para as espécies vegetais. A atividade antibacteriana in vitro foi observada por meio dos métodos de difusão em disco de papel, por hole- plate e por template, frente a Staphylococcus aureus, Bacillus subtilis, Escherichia colik, Enterococcus faecalis e Pseudomonas aeruginosa. A concentração inibitória mínima (CIM) foi determinada por meio do método de macrodiluições sucessivas em caldo. Os resultados obtidos apresentaram-se de acordo com o histórico farmacognóstico das drogas estudadas. Todas as bactérias foram inibidas pelos extratos, observando-se que as espécies S. aureus e B. subtilis mostraram, aparentemente, maior sensibilidade. A CIM variou, em relação a sensibilidade de cada espécie bacteriana estudada, de 312,5 æL/mL a 1250 æL/mL para os extratos vegetais e de 625 æL/mL a 2500 æL/mL para o enxaguatório bucal. São necessários estudos complementares para a confirmação da eficácia deste produto e sua utilização na prevenção de doenças periodontais.


In this study, herbs and hidroalcoholic extracts of Nasturtium officinale, Rosmarinus officinalis, Tabebuia impetiginosa, Plantago major and Achillea millefollium, were investigated for pharmacognostics analysis (flavonoids, alkaloids, tanins, saponins and antraquinones) and antibacterial activity (against Staphylococcus aureus, Bacillus subtilis, Escherichia coli, Enterococcus faecalis e Pseudomonas aeruginosa). In vitro antibacterial studies were carried by the disc diffusion method, hole plate and tamplate. The minimum inhibitory concentration (MIC) values were determinated by broth dilution method. The extracts were incorporated in mouthwash for periodontal diseases treatment. The pharmacognostics studies showed that herbs and extracts have substances groups described in literature. The antibacterial studies showed that extracts and mouthwash have some activity against all considered bacteria. S. aureus and B. subtilis showed, apparently, to be species more susceptible. Investigation of the MIC showed 312,5 æL/mL-1250 æL/mL for extracts and 625 æL/mL-2500 æL/mL for mouthwash. The studies served as initial quality control of the obtained cosmetic, enhancing anti-inflammatory (flavonoids) and/or antibacterial activities (tanins). However, new studies are necessary to confirm the cosmetic efficacy for treatment periodontal diseases.

In the present study, effects of pH (5.0, 7.0 and 9.0) and lead (1, 5, 10, 25, 50 and 100 microg mL(-1)) were investigated on uptake of lead, content of chlorophyll and nitrogen in Nasturtium officinale and Mentha aquatica. Total chlorophyll and nitrogen contents were adversely affected from Pb2+ concentrations dose dependently at each pH. The macrophytes were adversely affected by pH 5.0 or more than 9.0. After 12-days Pb2+ treatment, results showed that lead accumulation of macrophyte tissues was variable. According to the parts of the macrophytes, Pb2+ amounts were generally found at all tested pH levels and the metal concentrations for M. aquatica in the following order: root > stem > leaf and for N. officinale root > leaf > stem. Pb2+ concentrations in plant tissues (root, stem, leaf) in relation to pH were generally found for both macrophytes in following order: 7.0 > 9.0 > 5.0. Despite of the fact that high Pb2+ accumulation was observed in root tissues of the macrophytes, low metal accumulation was measured in the above-ground parts indicating low root-leaf translocation. The study indicated that uptake rate of Pb2+ and its toxicity on Chlorophyll and nitrogen contents in the macrophytes were dependent upon pH value of solutions.

ABSTRACT

El objetivo del trabajo fue realizar un estudio de foco relacionado con un caso de fasciolosis humana ocurrido en abril de 2002 en Loncopué, Neuquén, Argentina. La confirmación diagnóstica se efectuó en mayo de 2002 por el test de ELISA. En noviembre de 2002 se realizó un muestreo en el área rural donde se ubicaba la vivienda de la paciente, ya restablecida, y se le tomó una nueva muestra de sangre. El suero de la paciente continuó reactivo para antígenos de Fasciola hepatica. Se muestrearon plantas de berro para detectar metacercarias. Se recolectaron caracoles en cuatro canales de riego conectados a un canal principal. Los caracoles fueron trasladados vivos para su identificación, medición y examen de infección. Se recolectaron 35 muestras fecales de ganado de cría. No se observaron metacercarias en las hojas de berro examinadas (n=222). Se recolectaron 130 caracoles identificados como Lymnaea viatrix y 2 de 101 ejemplares (2%) estaban infectados con larvas de F. hepatica. Las prevalencias en el ganado adulto fueron: 100% (10/10) para caprinos, 82% (9/11) para ovinos y 86% (6/7) para bovinos. El número de huevos eliminados por las cabras (mediana = 20.7; Q1=6.2; Q3=34.5) y ovejas (4, 18, 13) infectadas, resultó mayor que el eliminado por vacas (0.3; 0.3; 1.7) (p<0.01). La práctica de control local no tuvo efecto aparente en este caso, por lo que deberían revisarse los calendarios de tratamiento y los antiparasitarios utilizados. Los resultados muestran que el ganado criado a pequeña escala por los pobladores debe incluirse en los programas de control. Se discute la posible importancia de la fasciolosis humana en Argentina.

ABSTRACT

Aquatic plants are well known in accumulating and in concentrating heavy metals. In this study, several physiological responses of aquatic vascular plant, Nasturtium officinale [watercress], which were elevated concentrations of copper, zinc and nickel have been investigated. It was found that Nasturtium officinale were able to accumulate both copper and zinc at upper levels, but was able to accumulate to nickel at low levels. The final goal of this work was to examine the copper, zinc and nickel uptake using this aquatic plant from their solutions
